Involvement of hyperpolarization-activated, cyclic nucleotide-gated cation channels in dorsal root ganglion in neuropathic pain

摘要

Dorsal root ganglion (DRG) neurons have peripheral terminalsin skin, muscle, and other peripheral tissues, andcentral terminals in the spinal cord dorsal horn. Hyperpolarization-activated current (Ih) of the hyperpolarizationactivated,cyclic nucleotide-gated (HCN) channels arepresent in the DRG. The genes encoding HCN channelshave four subtypes named HCN1 to HCN4. HCN channelsare permeable to both K+ and Na+. They underlie thedepolarization that modulates the rhythmic generations ofaction potentials (APs), contribute to the resting membranepotential, and modify the waveform of propagated synapticand generator potentials.Neuropathic pain is characterized by spontaneous pain,hyperalgesia and allodynia. After spinal nerve injury, thecell bodies of the primary sensory neurons in segmentalDRG become hyperexcitable, characterized for some neuronsby the presence of spontaneous firing (or ectopicdischarge). In the following, we summarize our observationson the role of HCN channels in DRG neurons in neuropathicpain.
