电针对大鼠脑内雌激素受体蛋白及其mRNA表达的影响

摘要

本文应用放射免疫分析(RIA)、RNA点杂交和Northernblot、单克隆抗体免疫组织化学和计算机图像处理技术研究电针对切除卵巢大鼠脑内雌激素受体(ER)蛋白和mRNA表达的影响。以探讨针刺作用的分子生物学机理。结果表明,切除卵巢可导致血雌二醇(E2)水平降低,动物脑内ER蛋白和mRNA的表达增强;电针实验穴位后,去卵巢大鼠血的E2含量明显增加,脑内ER蛋白和mRNA表达受到明显抑制。正常大鼠电针处理后,血E2水平和脑内ER表达均未见明显改变。上述结果提示,电针可提高去卵巢大鼠体内雌激素水平,使脑内ER表达发生改变,这种作用是一种涉及机体某些基因表达改变的长时程效应,初步看来似乎是针刺调整下丘脑-垂体-卵巢轴异常功能的作用机制之一。

Abstract

In order to show some light on the possible molecular mechanism of the effects ofacupuncture on the hypothalamic-pituitary-ovarian axial function,radioimmunoassay(RIA), RNA dot blot and Northern blot, monoclonal antibody immuno-histochemistryand computer image analysis technique were used to study the effects ofelectroacupuncture(EA) on the blood level of estradiol (E2) and the expression ofestrogen receptor protein and mRNA in the ovariectomized rat brain. The results show thatovariectomy induced a decrease of blood E2 level and increase of the expression ofestrogen receptor protein and mRNA in the brain. All these effects could be remarkablyreversed by previous EA treatment of ovariectomized rats. EA in intact rat, however, hadno effects on blood E2 and expression of estrogen receptor.
