用高效液相色谱—电化学检测法快速、灵敏分析生物样本中的单胺类递质及其代谢产物

摘要

该实验用反相离子对色谱-电化学检测分析法,同时分离并测定生物样本中结构极为相似的单胺类递质及其代谢产物。实验采用等比洗脱,在30min内至少可测定九个这类化合物。生物样品的预处理简单,因而平均回收率能达到90%以上。十个化合物(包括3,4-二羟基苄胺)保留时间变异系数为0.73±0.18%(均值±标准差,下同);峰面积变异系数为1.11±0.45%;在50—100pg至40ng范围内,相夫系数为0.996±0.006。本文对我们所用的色谱分析条件进行了讨论。

Rapid and sensitive assay of monoamine iransmitters and their metabolites in biological samples using high performance liquid chromatography with electrochemical detection

Abstract

In the present experiment, monoamine transmitters and their metabolites, which had very analogous structure, were simultaneously separated and measured by reversed-phase ion-pair chromatography with electrochemical detection. The
isocratic elution was used and this method was able to determine nine compounds
among these compounds, at least, at one run during 30 min. The preparation of
biologic samples was simple, so the mean recovery could reach over 9000. The
mean coefficient of variation in retention time of ten compounds (include 3,4-dihydroxybenzylamine) was 0.73%±0.18% (mean1 S.D.), and the mean coefficient
of variation in peak area was 1.11%±0.45% (mean1 S.D.). In the range of 50-100pg to 40ng, the linear relation between the amounts and the peak areas was
accurate (r=0.996±0.006, mean±S.D.). The chromatographic conditions performed in the present assay were discussed.
