GABA介导杏仁外侧核对皮层A Ⅰ区神经元声反应的抑制:在体微电泳研究

摘要

在SD大鼠上应用多管微电极方法,观察微电泳GABA及其受体的拮抗剂或激动剂对杏仁外侧核(LA)抑制皮层A-Ⅰ神经元声反应效应的影响.结果显示,电泳GABA能抑制皮层A-Ⅰ区神经元的电活动,电泳GABAA受体拮抗剂bicuculline(BIC)则能易化其反应;电刺激LA能抑制皮层A I区听神经元声反应,电泳GABA产生类似于刺激LA的抑制效应;LA对皮层A-Ⅰ区神经元的抑制效应能被BIC所翻转,而不能被甘氨酸受体拮抗剂strychnine所翻转,电泳GABAB型受体激动剂bac1ofen对神经元声反应无影响.上述结果表明,GABA可能是介导LA抑制皮层A Ⅰ区神经元声反应的最终递质,并且是通过GABAA受体起作用的。

GABA mediaties the inhibitory effect of lateral amygdaloid nucleus stimulation on the acoustic response of neurons in A Ⅰ cortex:An in vivo microiontophoretic study

Abstract

Experiments were performed on Sprague Dawley rats with multibarrel microelectrode technique. The effects of acoustic response of A-Ⅰ cortex neurons produced by electrical stimulation of lateral amygdaloid nucleus (LA) and the influence of GABA were observed. Experimental results showed that iontophoretic administration of GABA caused a pronounced inhibition of the electrical activity of A-Ⅰ neurons. Blockade of GABAA with bicuculline (BIC) facilitated the acoustic response. The acoustic response of A-Ⅰ neurons was inhibited when the LA was stimulated. Iontophoretic application of GABA resulted in a similar inhibitory effect as that of LA stimulation. Blockade of GABAA with bicuculline reversed the inhibitory effect of LA stimulation on the acoustic response of A-Ⅰ neurons. In contrast, application of strychnine, a glycine receptor antagonist, could not reverse the inhibitory effect of LA. Baclofen, a GABAB agonist, did not affect the acoustic response of the auditory neurons. These results indicate that GABA is the ultimate transmitter which mediates the LA stimulation-induced inhibition of the acoustic response of A-Ⅰ neurons in rats, possibly via the GABAAreceptor
