肿瘤坏死因子对大鼠中脑神经干细胞分化和寡突胶质细胞增殖的影响

摘要
为观察肿瘤坏死因子对神经干细胞(NS Cs)分化的影响,本研究应用体外扩增的新生大鼠中脑NSCs,使用免疫组织化学技术,观察了肿瘤坏死因子一。(TNFa)对NSC、分化及其后代细胞的影响。结果显示:(1) TNFa可提高中脑NS Cs后代中神经元和寡突胶质细胞所占的比例;(2) TNF-。可明显诱导由NS Cs分化的寡突胶质细胞增殖,但对星形胶质细胞的增殖作用不明显。上述观察结果提示TNF-。对NS Cs的应用具有重要影响。

Abstract
To observe the influence of tumor necrosis factora(TNF-a) on differentiation of rat mesencephalic neural stem cells(NS Cs),the numbers of neurons,astrocytes and oligodendrocytes generated from NSCs were analyzed after differentiation for 3 days by using i m munocytoche mistry technique.The
results show that:(1)TNF-a enhanced the proportions of neurons and endrocytes in progeny of NS Cs;and(2) TNF-a induced the proliferation of oligodendrocytes derived from NSCs,but the prolifer ation of astrocytes was not influenced by TNF-a.We conclude that the TNF-a could influence the application of NSCs.
