高盐负荷对大鼠血管紧张素Ⅱ(A Ⅱ)受体mRNA表达的影响

摘要

实验采用逆转录—聚合酶链式反应(RT-PCR)技术,比较了12周龄卒中型自发性高
血压大鼠(SHRsp)和正常血压大鼠(WKY)主动脉平滑肌、心室肌和肾上腺等组织的血管紧张
素d受体的la亚型(ATla)和lb亚型(ATlb) mRNA的表达,同时检测了高盐负荷对两种大鼠
ATla和ATlb受体mRNA表达的影响。结果显示:(1)在SHRsp, ATla和AT 1b mRNA在肾上
腺的表达明显高于W KY大鼠(分别高 176%和15700),其它组织的表达均低于WKY大鼠(在
主动脉平滑肌分别低10%和2300,在心室肌分别低23%和4000)0 (2)高盐负荷后,WKY大鼠
肾上腺的ATla和ATlb及心室肌的ATlb mRNA的表达显著增加(在肾上腺分别高167%和
4010o,在心室肌高6200),主动脉平滑肌ATla和ATlb及心室肌AT la mRNA的表达未见明显
改变。(3)高盐负荷后,SHRsp主动脉平滑肌的ATlb及心室肌的ATla和ATlb mRNA的表达
均显著增高(在主动脉平滑肌高9000,在心室肌分别高590%和20000),肾上腺的AT la mRNA
的表达明显下降(下降5800),主动脉平滑肌的ATla及肾上腺的AT 1b mRNA的表达未见明显
改变。初步结果提示:高盐负荷时,ATIa和ATlb受体均参与了高血压的发生和发展;主动脉平滑肌AT l b受体以及心室肌ATla和ATlb受体的改变有可能引起血管平滑肌细胞肥大、血管结构改变和心肌肥厚;SHRsp和WKY大鼠某些组织之间的ATla和ATlb受体mRNA的表达,在
有或无外界刺激因素的情况下均存在一定差异。

EFFECTS OF HIGH SALT-LOADING ON THE REGULATION OF ANGIOTENSIN II RECEPTOR mRNA EXPRESSION

Abstract

In the present study,the angiotensin 11 receptor subtype I一a (ATla) and I一b(AT 1b)mRNA levels in aortic smooth muscle(ASM),ventricular myocardium
(VM) and adrenal from 12一week一old stroke-prone spontaneously hypertensive rats (SHRsp) and age-matched Wistar-Kyoto (WKY) rats with normal diet (control) and high salt-loading were examined by reverse transcriptase- polymerase chain reaction (RT-PCR).The results showed that:(1)The AT] a and ATlb mRNA levels in ASM and VM from SHRsp were lower than those from WKY rats(in ASM,10%and 23%,while in VM,23%and 4 0%lower,respectively).In contrast,both AT la and AT] b mRNA levels in adrenal from SHRsp were higher(176%and 157%,respective1y).(2)In the WKY rats with high salt-loading,the AT Ia and AT 1b mRNA levels in adrenal,as well as AT]b mRNA level in VM,increased significantly,as compared with the control (in adrenal,167%and 401%,while in VM,62%}.However,the AT 1 a and AT I b mRNA levels in ASM,as well as AT] a mRNA level in VM,showed no obvious change.(3)In SHRsp with high salt-loading,the AT] b mRNA level in ASM,as well as AT l a and AT l b mRNA levels in VM,increased markedly (in ASM, 90%,while in VM,590%and 200%);whereas the AT 1 a mRNA level in adrenal decreased significantly(58%).There was little influence on the regulation of AT l a (in ASM)and ATIb (in adrenal)receptor gene expression after high salt-loading. The results suggest that AT l a and AT 1 b receptors may be involved in the pathogenesis of saltinduced hypertension. The up-regulation of AT l b receptors in ASM may induce the re
modeling of arterial wall,while that of AT la and AT 1b receptors in VM might contribute to ventricular hypertrophy in hypertension. Furthermore,there are certain differences between SHRsp and WKY rats with respect to the regulation of ATIa and AT I b receptor gene expression with or without external stimulation.
