内皮素对培养心肌细胞内游离钙浓度的作用

摘要
以Fura-2/AM荧光指示剂负载检测心肌细胞内游离钙浓度([Ca~(2+)]_(i))的变化,探讨内皮素-1(ET-1)对[Ca~(2)]_(i)的作用及其机制。结果显示:ET-1引起心肌细胞9Ca~(2+)]_(i)升高有两个时相,瞬时相和持续相。ET-1诱导的瞬时相[Ca~(2+)]_(i)升高呈浓度依赖性,预先用ET_(A)特异性受体阻断剂BQ123处理,可阻断ET-1引起的[Ca~(2+)]_(i)升高。

Abstract
In this present study, the effects of ET-1 on intracellular free calcium concentration ([Ca~(2+)]_(i)) and the underlying mechanisms were investigated in cultured neonatal rat myocardail cells loaded with fura-2/AM. The results are as follows. ET-1 induced an increase of [Ca~(2+)]_(i) in a dose-dependent manner, which consisted of transient and sustained phase. BQ123, a selective ET_(A) receptor antagonist, blocked the ET-1 induced [Ca~(2+)]_(i) responses, suggesting that these responses were mediated by ET_(A) receptors.
