HAP转录因子与DNA结合的活力受细胞内氧化还原系统的调节

摘要

在酵母hap5突变株的细胞中,用酵母单杂交系统从水稻cDNA-GAL4表达文库中筛选出的阳性克隆是编码谷胱甘肽氧还蛋白的cDNA,提示细胞内的氧化还原系统可能作用于HAP蛋白,从而对CCAAT盒的结合活力起调节作用。对HAP3亚基分子中半胱氨酸残基的突变实验结果支持上述推测。

DNA binding activity of the transcription factor HAP is regulated by cellular redox

Abstract

By using a yeast one-hybrid system, the authors tried to isolate the counterpart of the yeast HAP5 gene from rice cDNA-Gal4 fusion library constructed in a E. coli-yeast shuttle vector pPC86. However, the authors identified a cDNA encoding glutaredoxin. The results indicate that the cellular redox environment of yeast cell may be a factor regulating the CCAAT-box-binding activity of the HAP3 subunit. The results of the HAP3 mutants in which the highly conserved cysteine residues at positions 68 andn 72 had been converted to serine support the above suggestion.
