H_(2)O_(2)和·OH及其清除剂对大麦叶片液泡膜微囊质子转运活性的影响

摘要

大麦叶片液泡膜微囊经H_(2)O_(2)和·OH处理后,H~(+)-ATPase和H~(+)-PPase水解活性和泵运质子活性下降,H~(+)-PPase对H_(2)O_(2)更敏感一些。同时加入与H_(2)O_(2)和·OH相同浓度的抗坏血酸(AsA)或甘露醇可显著恢复两种酶的活性和质子转运活性。H_(2)O_(2)和·OH处理后,H~(+)-ATPase米氏常数变大,H~(+)-PPase的最大反应速度减小。

Effects of H_(2)O_(2), ·OH and their scavengers on the activities of H~(+)-transport in the tonoplast vesicles isolated from barley leaves

Abstract

Both hydrolytic and H~(+)-transport activities of H~(+)-ATPase and H~(+)-PPase in the tonoplast vesicles isolated from barley leaves were lowered by H_(2)O_(2) and ·OH treatments. The hydrolytic activity of H~(+)-PPase was more sensitive to H_(2)O_(2) than that of H~(+)-ATPase. The depressed hydrolytic and H~(+)-transport activities of these two enzymes were recovered significantly by the addition of AsA, mannitol at the same concentration as H_(2)O_(2) and ·OH. K_(m) value of H~(+)-ATPase was increased and V_(max) of H~(+)-PPase decreased by treatments with H_(2)O_(2) and ·OH.
