Wnt信号通路关键基因#beta#-catenin在RA诱导的P19胚胎性癌细胞神经分化过程中的亚细胞分布

摘要

以Wnt信号通路关键成员#beta#-catenin是否定位在细胞核中作为考察Wnt信号是否能传递到细胞核内调控下游基因活性的指标,分析了Wnt信号在P19细胞神经分化过程中的活性。#beta#-catenin在RA诱导的P19细胞分化的第2到第4天存在核定位。在P19神经分化过程中,Wnt活性可能发生在这一阶段。同时,在神经突的形成、伸长及神经纤维集束过程中,#beta#-catenin在神经突起及神经纤维上也存在特异性分布,提示在此过程中#beta#-catenin可能也行使了功能。

Abstract

Wnt signals have been shown to play an important role in the development of the central nervous system (CNS) of mouse. In our previous work, it was demonstrated that Wnt signal could initiate differentiation of P19 EC cells. In the present investigation, it was examined with RT-PCR whether expression of #beta#-catenin, a downstream gene of Wnt in its signal transduction pathway, is regulated.
