杜氏盐藻(Dunaliella salina)盐适应相关蛋白

摘要

观察不同盐浓度培养液中生长的杜氏盐藻可溶性蛋白SDS-PAGE图谱,发现高盐与低盐相比,其51kD和63kD蛋白含量高,而23kD的蛋白含量则低。高渗骤变后,51kD蛋白的含量减少,而23kD蛋白的含量增加两倍或两倍以上,骤变23h后含量增加已非常明显;低渗骤变后24h,51kD和23kD蛋白的含量变化不明显。另外,有一分子量约为26kD的蛋白在高盐下易降解。分析了这些蛋白与杜氏盐藻渗透调节的可能关系。

Proteins associated with adaptation of the green alga Dunaliella salina to NaCl stress

Abstract

SDS-PAGE profiles of cell extracts from the green alga, Dunaliella salina, grown in different salinities revealed that the amount of the 51 kD, 23 kD and 63 kD proteins were more abundant in the alga cells when the concentration of the NaCl in the culture medium was 1 mol/L than when it was 3 mol/L. In contrast, the 23 kD protein was more aboudant in cells grown in culture medium with NaCl 3 mol/L; Its relative amount in cells grown in culture medium with NaCl 3 mol/L or more was about two folds higher than that of cells in culture medium with NaCl 1 mol/L or less. Furthermore, when the cells grown in culture medium with NaCl 1 mol/L were subjected to drastic hyperosmotic shock (NaCl 1 mol/L→3 mol/L), the amount of 23 kD protein increased, while the amount of 51 kD protein decreased. No obvious changes of the amount of the two proteins were observed when the cells grown in culture medium with NaCl 3 mol/L were subjected to hypoosmotic shock→NaCl 3 mol/L→1 mol/L) even after 24h treatment. In addition, an unstable 26 kD protein was easily digested. The possible relationship of these proteins with osmoregulation were analyzed as well.
