绿豆上胚轴细胞中BR的胶体金免疫电镜定位

摘要

利用葡萄球菌A蛋白与胶体金连接的复合物为探针的免疫电镜定位技术对绿豆上胚轴细胞中BR定位的结果表明,在用抗BR抗体处理的超薄切片中,叶绿体、核仁和液泡内有大量的金颗粒标记,细胞膜和淀粉粒也有金颗粒标记,但细胞壁中没有观察到金颗粒。在不用EDC固定的切片中,金颗粒标记密度非常低,而在用正常兔血清处理的切片中,所有细胞器内几乎没有金颗粒。

Immunogold electron microscopy localization of brassinolide in epicotyl cells of mung bean

Abstract

With immunogold (PA-Au complexes) electron microscopy the localization of brassinolide (BR) in epicotyl cells of mung bean (Phaseolus radiatus L.) has been studied. The results showed that, in thin sections treated with anti-BR, choroplasts, nucleoti, vacuoles, starch grains and cell membranes were labelled by gold particles, with choroplasts, nucleoti and vacuoles being labelled more heavity. However, no gold particles were found in cell walls. The sections with no EDC fixation contained very few gold particles, and those treated with preimmune rabbit serum contained much fewer gold particles.
