红豆草抗羟脯氨酸变异系原生质体培养后愈伤组织的抗性特征

摘要

通过NaN_(3)诱变得到的红豆草抗羟脯氨酸(Hyp)变异系,在酶液中游离原生质体进行培养,获得再生植株。 在含不同浓度NaCl、羟脯氨酸或PEG的MS培养基上,原生质体来源抗性愈伤组织中的游离脯氨酸含量在1wk之内均急剧增加,随后开始下降, 3wk后接近正常水平。随着胁迫程度的提高,抗性愈伤组织中游离脯氨酸含量呈递增趋势,生长速度呈递减趋势。

Resistant characteristics of the hydroxyproline resistant cell line in Onobrychis viciaefolia after protoplast culture

Abstract

The hydroxyproline resistant cell line of Onobrychis viciaefolia had been selected through NaN_(3) mutagenesis. Protoplast-derived plantlets of the resistant cell line were regenerated in four months and were found to retain strong hydroxyproline resistance. After the calli of the resistant cell line derived from protoplasts were treated with different concentrations of NaCl,Hyp or PEG, the free proline content increased steeply in the first week and descended to normal level in three weeks. In contrast to this, proline accumulation in the wild tissue was not found at low selection level it could stand. Along with the enhancement of stress, the free proline concentration in calli of the resistant cell line increased, while the relative growth rate descended. When the resistant cell line was transferred from selection medium onto normal MS medium, its free proline content declined to the normal level in six days. It suggested that free proline accumulation was related to osmotic regulation and was the major way to prevent water loss of the resistant cell line under water stress. Total amino acid analysis revealed that methionine content of the regenerated plantlet leaves derived from hydroxyproline-resistant protoplasts was twice as high as the original type.
