谷子胚性愈伤组织粘液提取物对原生质体培养的影响

摘要

将谷子胚性愈伤组织粘液提取物添加到谷子原生质体培养基中,其对原生质体培养的影响表明该提取物有助于原生质体形成细胞壁;并且该类有粘液分泌的愈伤组织的原生质体游离所需的酶液浓度低、处理时间短。由原生质体形成完整细胞的数量在一定范围内与谷子原生质体培养的植板率相对应;通过增加形成完整细胞的数量可较大幅度地提高原生质体培养的植板率。

The effect of mucilaginous substances extracted from foxtail millet embryogenic callus cultures on its protoplast cultures

Abstract

Embryogenic callus cultures of cereal crops are usually accompanied by the excretion of mucilaginous substances, but the composition and function of these substances are yet to be analyzed. In this paper, the authors reported the isolation of protoplasts from embryogenic calli with mucilage by brief enzyme treatment,and the protoplast isolation was greatly facilitated by coupling moderate mechanic application. The authors added the mucilage extract from embryogenic callus of foxtail millet (Setaria italica L. Beauv.)to the media for culture of its protoplasts,to assess the effect of the extract on protoplast culture.The results showed that the addition of the extract promoted cell wall formation by protoplasts.Within the range of test,the number of the intact cells formed from protoplasts was correlated with the plating efficiency of protoplast culture,which was greatly enhanced by the addition of the mucilage extract to the protoplast culture media.
