磷脂酶A_(2)介导肿瘤坏死因子诱发的中性粒细胞趋化和粘附增强

摘要
肿瘤坏死因子(TNF)处理60min后, 外周中性粒细胞(PMN)对白细胞趋化因子(FMLP)趋化和玻璃珠粘附性均显著增加, 同时伴有血栓素B_(2)(TXB_(2)释放增多, 前 列环素(6-keto-PGF_(1a)不变。PLA_(2)代谢产物花生四烯酸(AA)处理60min 后, PMN趋化和粘附性也显著增加, 并伴有TXB_(2)释放增多。PLA_(2) 抑制剂 PBPB可大部阻断TNF诱导的PMN趋化和粘附增加, 而环氧化物酸抑制剂氯苯甲酰吲哚乙酸(IND)对TNF诱导的上述PMN功能无显著影响。

Abstract
TNF(100ng/ml)given in vitro for 60 min, causes an obvious enhancement of neutrophil chemotaxis towards FMLP, and increased activity of adhesion toglass beads by stimulated release of TXB_(2) from PMN other than 6-keto-PGF_(1a). Similer neutrophil activation responces could also be promoted by in vitro addition of arachidonic acid (1 #mu#mol/ml)for 60 min. It was further found that PLA_(2)inhibitor, 4-bromophencyl bromide (5#mu#mol/ml), greatly inhibite the TNF induced responces, while cycloxgenase inhibitor indomethathin (85nmol/ml) had no effect on the activation of TNF.
