紫竹梅雄蕊毛发育过程中胞间连丝通透能力的动态变化

摘要

紫竹梅开放花和花蕾IV期胞间连丝允许通过物质的分子量不超过1000D, 衰老花可允许FITC-insulin A链(2921D)通过, 而花蕾I期可允许FITC-dextran (4400D)通过。说明 胞间连丝允许通过物质的分子量极限不是固定不变的, 它随组织、细胞的发育进程而改变。

Changes in plasmodesmatal permeability of the staminal hairs of Setcreasea purpurea during development

Abstract

A range of water-soluble fluorescent dyes of known molecular weights were injected into staminal hairs of Setcreasea purpurea at different developmental stages by pressure with Opton Microinjection System. Fluorescein isothiocyanate (FITC) was conjugated to (Ala)6 and Leu-(Arg)2-Ala -Ser-Val-Ala. and purified. LRB and FITC-dextran were made up as 1mmol/L solution, and the movement of the fluorecein probes was observed under the fluorescence microscope (Olympus RFL) 5min after the beginning of dye injection. Only where dyes was seen in the neighboring cells was it cored as dye movement. The results show that plasmodesmatal permeability changes greatly with the development of staminal hairs. the molecular exclusion limit of plasmodemata in bud IV is less than 1000D which is similar to that found in previous works.
