玉米叶绿体atpB基因表达产物的特性及其抗体对CF_(1)功能的影响

摘要

利用大肠杆菌获得玉米叶绿体atpB融合基因及非融合基因的高效表达。从大肠杆菌中 部分分离纯化得到的这些atpB基因表达产物都表现出微弱的水解ATP的活力。由其中一种融合蛋白#beta#_(1)制备得到的抗血清对菠菜叶绿体偶联因子的光合磷酸化功能影响不明显, 对游离CF_(1)的Ca~(2+)-ATP酶活力也无明显影响, 但却能明显地促进它的Mg~(2+)-ATP酶活力。

Properties of maize chloroplast atpB gene expression products and effects of its antibody on the functions of CF_(1)

Abstract

Maize chloroplast atpB fusion and non-fusion genes were over-expressed in Escherichia coli. These expression products in E. coli were partially purified. They all exhibited very weak ATP-hydrolyzing activities. Anti-serum against fusion protein #beta#_(1) had no obvious effects on the photophosphorylation of spinach chloroplasts and Ca~(2+)-ATPase activity of soluble CF_(1) (Tables 4,5). However, it could increase the activity of soluble CF_(1) Mg~(2+)-ATPase stimulated by methanol. This indicates that soluble CF_(1) stimulated by methanol may show both activies of Mg~(2+)-and Ca~(2+)-ATPase, but their conformations at hydrolyzing ATP may be different.
