八肽胆囊收缩素拮抗NDAP对大鼠脊髓钙调蛋白的作用

摘要

八肽胆囊收缩素拮抗(CCK-8)可通过CCK受体介导增加大鼠脊髓背柱突触小体钙调蛋白(CaM)活性, 并能拮抗NDAP对CaM活性的抑制作用, CCK-8的这一作用可能是其抗阿片作用的机理之一 。

Cholecystokinin octapeptide antagonized the effect of NDAP on CaM in rat dorsal spinal synaptosome

Abstract

The effects and interaction of CCK-8 and k opiate agonist NDAO on CaM activity in rat dorsal spinal cord synaptosomes are observed. The result indicated that 10nmol/L=1umol/L NDAO produced dose-dependent decrease in CaM activity of rat spinal synaptosomes. This suppressive effect was completely blocked by specific k receptor antagonist Nor-BNI at 1 umol/L. In constrast CCK-8 induced a significant increase in CaM activity, and was capable of antagonizing the suppressive effect induced by NDAP. Specific CCK-B receptor antagonist L365,260 could partially block the effects of CCK-8 increased CaM activity at 1 umol/L. The above results suggest that CCK-8 can antagonize the suppressive effect of NDAP on CaM activity in rat spinal synaptosomes via activition of CCK_B receptor, an effect that may constituti one of the mechanisms for the anti-opioid activity of CCK-8.
