泛素化对经典NF-κB信号通路调控机制的研究进展

摘要

NF-κB信号通路是炎症和免疫应答的关键调控通路,其激活可促进炎症因子产生和抑制细胞凋亡。在经典NF-κB信号通路中,泛素化修饰起着重要的调节作用,其中赖氨酸残基48、63 位点(K48、K63)以及甲硫氨酸1 位点(Met1)结合形成的泛素链与NF-κB通路有着更为紧密的联系。本文旨在总结K48、K63 和Met1 连接的泛素化在经典NF-κB信号通路中的作用,对深入了解这些泛素连接类型的作用机制以及揭示其对NF-κB通路的分子调控机制有重要意义,并为炎症和免疫相关疾病的治疗提供新的靶点和策略。

Research progress on the regulatory mechanism of ubiquitination on canonical NF-κB signaling pathway

Abstract

NF-κB signaling pathway is a key regulatory pathway of inflammation and immune response, and its activation can promote the production of inflammatory factors and inhibit apoptosis. In the canonical NF-κB signaling pathway, ubiquitin modification plays an important regulatory role, in which the ubiquitin chain formed by lysine residues 48, 63 (K48, K63) and methionine 1 (Met1) is more closely related to the NF-κB pathway. This paper aims to summarize the role of K48, K63 and Met1-linked ubiquitination in the canonical NF-κB signaling pathway, which is of great significance to further understand the mechanism of these ubiquitin-linked types and reveal their molecular regulation mechanism on NF-κB pathway, and provide new targets and strategies for the treatment of inflammatory and immune-related diseases.
