细胞间对话与肝细胞癌进展

摘要

恶性增殖的肝癌细胞可以通过多种方式感知机体信号,以利于肿瘤自身的增殖、侵袭和转移等。细胞间对话(Crosstalk)是肝癌细胞感知机体信号的重要途径之一。在肝细胞癌(hepatocellular carcinoma, HCC)组织中,HCC细胞可以与肝星状细胞(hepatic stellate cells, HSCs)、肿瘤相关巨噬细胞(tumour-associated macrophages, TAMs)和免疫细胞等Crosstalk,进而调控肿瘤的增殖、侵袭和转移等过程。Crosstalk 具有多种方式,每种方式具有不同的功能特点,其分子机制主要包括表达CD24、CD47 等特异性受体,调控免疫细胞功能;分泌转化生长因子-β (transforming growth factor-β, TGF-β)、血小板衍生生长因子(platelet-derived growth factor, PDGF)等细胞因子,调控细胞功能;激活Wnt/β-catenin、Hedgehog 通路,调控细胞生长、增殖等。深入理解Crosstalk 的作用和机制,有助于揭示HCC的发病机制,并为开发新的治疗策略提供思路和靶点。本文综述Crosstalk与HCC进展的关系,为进一步研究提供参考和启示。

Crosstalk and the progression of hepatocellular carcinoma

Abstract

Malignant proliferating liver cancer cells possess the ability to detect and respond to various body signals, thereby facilitating tumor growth, invasion, and metastasis. One crucial mechanism through which hepatocellular carcinoma (HCC) cells interpret these signals is crosstalk. Within liver cancer tissues, cancer cells engage in communication with hepatic stellate cells (HSCs), tumorassociated macrophages (TAMs), and immune cells. This interaction plays a pivotal role in regulating the proliferation, invasion, and metastasis of HCC cells. Crosstalk occurs in multiple ways, each characterized by distinct functions. Its molecular mechanisms primarily involve regulating immune cell functions through the expression of specific receptors, such as CD24 and CD47, modulating cell functions by secreting cytokines like transforming growth factor-β (TGF-β) and platelet-derived growth factor (PDGF), and mediating cell growth and proliferation by activating pathways such as Wnt/β-catenin and Hedgehog. A comprehensive understanding of the mechanisms and interactions within crosstalk is essential for unraveling the pathogenesis of HCC. It also opens up new avenues for the development of innovative therapeutic strategies. This article reviews the relationship between crosstalk and the progression of HCC, offering insights and inspiration for future research.
