自发性高血压大鼠心肌肥厚和心肌MAPK、AngⅡ的关系

摘要

放免法测定自发性高血压大鼠(SHR)血浆及心肌血管紧张素Ⅱ(AngⅡ)含量,凝胶内磷酸化法测定心肌丝裂素活化蛋白激酶活性(MAPK),以心脏重/体重表示心肌肥厚程度。结果表明:与4个月的WKY大鼠比较,4个月的SHR血浆和心肌组织AngⅡ及心肌MAPK活性分别增加了218.6%、101.2%和107.0%,心肌肥大程度严重,其中MAPK活性与心肌肥大程度呈明显正相关。提示4个月SHR心肌肥厚可能是由MAPK介导的,AngⅡ可能参与了MAPK活性升高。与4个月SHR比较,15个月SHR心肌MAPK活性降低29.7%,血浆及心肌AngⅡ分别降低17.2%和31.3%。

Abstract

Mitogen-activated protein kinases (MAPKs) are thought to be critical components insignal transduction pathways in regulation of cell growth and differentiation. The purposeof the present investigation is to study possible involvement of MAPKs in the progress ofcardiac hypertrophy in spontaneously hypertensive rats (SHRs) and effects of age onAngiotensin Ⅱ (Ang Ⅱ), MAPK activity and cardiac hypertrophy. The animals weredivided into thee groups: 4 months old WKY rats (n=8), 4 month old SHRs (n=8)and 15 month old SHRs (n=6). Ratio of heart to body weight was measured. Ang Ⅱwas determined by RIA. MAPK activity in cardiac tissue was assayed by the "in-gel"myelin basic protein phosphorylation. The results show that in comparison with 4 monthold WKY rats, Ang Ⅱ in plasma and cardiac tissues were elevated (216.4%, P<0.01; 101.2%, P < 0.01 ) in 4 months old SHRs, while the MAPK activity wasincreased 107.0% (P < 0.01 ) with a parallel cardiac hypertrophy (P<0.01).
