α-黑色素细胞刺激素对白细胞介素-1β发热的解热机理研究

摘要

本研究观察了α-黑色素细胞刺激素(α-MSH)对家兔白细胞介素-1β(IL-1β)发热效应及下丘脑组织腺苷环-磷酸(cAMP)含量的影响;同时观察了下丘脑组织体外培养过程中,α-MSH对IL-1β刺激下丘脑释放cAMP的影响。结果显示:α-MSH能显著降低IL-1β引起的体温升高(P<0.05);同时抑制下丘脑组织cAMP含量的增高(P<0.01)。IL-1β与下丘脑组织培养,其上清液中cAMP含量明显升高(P<0.01);而预先加入α-MSH,再加同量IL-1β培养,则培养上清液的cAMP含量接近对照组。体内外实验表明,α-MSH抑制中枢发热介质cAMP,的生成可能是其解热的重要机制之一.

Abstract

The effect of α- melanocyte-sitmulating hormone (α-MSH ) on IL-1βinduced feverand release of hypothalaminc cAMP was observed in the present work. The results showedthat α-MSH markedly suppressed febrile response (P<0.05) and decreased the cAMPcontent in hgpothalamus in response to icy injection of IL-1β (P<0.01).The cAMPcontent of the medium in which the hypothalamic was incubated for 40 min in thepresence of IL-1βwas significantly increased (P<0.01). However, this increase actioncould be prevented by a pretreatment with α-MSH. The results indicate that the inhibitoryeffect of α-MSH on the enhanced synthesis and release of cAMP in hypothalamus exposedto IL-1β mingt be one of the mechanisms of suppressing fever induced by IL-1β.
