胰岛素样生长因子II对大鼠离体培养黄体细胞孕酮生成的影响

摘要

实验观察了胰岛素样生长因子B (IGF- Ⅱ)对大鼠离体培养黄体细胞孕酮生成的影响,并对其作用机制进行了探讨。结果显示,IGF- Ⅱ能显著地促进大鼠离体培养黄体细胞孕酮生成并呈剂量一效应关系,同时还能促进~(3)H一亮氨酸(3H-leu)掺入黄体细胞蛋白质的合成,促进~(3)H一胸腺啥吮(gH-TdR)掺入DNA的合成,而上述效应分别被放线菌酮(CYX)和放线菌素D (Act D)所抑制。此外,IGF- B对大鼠离体黄体细胞内源性cAMP和hCG诱导的CAMP含量均无影响,这提示IGF- B对大鼠黄体细胞孕酮生成的促进作用与其增强黄体细胞蛋白质与DNA的合成有关,而其跨膜信息传递似不通过。AMP系统。

EFFECT OF IGF-II ON PROGESTERONE PRODUCTION BY CULTURED RAT LUTEAL CELLS IN VITRO

Abstract

It has been reported recently that IGF- Ⅱ( plays an important role in the regulation of ovarian function. The aim of this study is to investigate whether IGF-Ⅱ( can affect progesterone production by cultured rat luteal cells. The results showed that IGF- Ⅱ(could significantly stimulate basal progesterone production in a dose-dependant manner without altering the cAMP content. IGF- Ⅱ could enhance 3H-leu incorporation into protein and 3H-TdR incorporation into DNA of the luteal cells,but these effects could be
inhibited by CYX and Act D. The above results suggest that stimulating effect of IGFⅡ( on progesterone production by rat luteal cells may be related to enhanced protein and DNA synthesis.
