氟哌利多增强电针镇痛时大鼠脑内微透析液中单胺类递质的变化

摘要

该文采用大鼠脑内微透析及高效液相色谱电化学联检技术,观察多巴胺(DA)受体拮抗剂氟哌利多(DRO)加强电针(EA)镇痛时大鼠脑内微透析液中单胺类递质的变化,从而探讨药物加强针刺镇痛的机制。结果显示,EA后DA及其代谢产物高香草酸(HVA)在透析液中含量增加(P<0. 05或P<0. 01), DRO与EA合用(DRO+EA)后,两者的含量较单用EA又有明显增加
(P<0. 05和P<0. 01 )。去甲肾上腺素(NE)含量在EA和DRO+ EA后均明显减少((P<0. 01) ,
但DRO + EA组与EA组之间无统计学差异((P>0. 05) 0 5-)t}色胺(5-HT)及其代谢产物5-)'叫噪乙酸(5-HIAA)在EA后显著增加,DRO+EA则使5-HT和5-HIAA进一步增加。以上结果提
示,DRO与EA合用后,不仅仅是通过拮抗DA,而且通过协调脑内其它单胺类递质的活动,从
而加强针刺镇痛。

ALTERATION OF MONOAMINE CONTENTS IN MICRODIALYSATE FOLLOWING DROPERIDOL ENHANCED ELECTROACUPUNCTURE

Abstract

The monoamine contents in microdialysate from rat brain before and after droperidol (DRO),a dopamine (DA) antagonist,enhanced electroacupuneture-induced analgesia (EA) were measured using techniques of in;二。microdialysis and high performance liquid chromatography with electrochemical detection (HPLC-EC),so as to investigate the monoamine mechanism of the potentiating effect of DRO on acupuncture analgesia (AA).The results showed that the levels of DA and its metabolites homovanillic acid (HVA) in perfusate increased after EA(P<0. 05 or 0. 0l),and further increased after DRO+EA (P<0. 05 and 0. 0l,vs EA treatment).A significant decrease in norepinephrine (NE) content after EA (P<0. 0l)was observed,but no further change after DRO+EA (P>O. 05,vs EA treatment).The contents of 5-hydroxytryptamine (5-HT) and its metabolite 5一hydroxyindole acetic acid(5一HIAA)we elevated after EA treatment,and a further elevation in 5-HT and 5-HIAA contents induced by DRO +EA was noted. The results in the present study suggest that DRO potentiated AA not
only via antagonizing the activity of dopaminergic system,but also by coordinating the activities of other monoamines in the brain.
