大鼠下丘脑离体脑薄片视上核神经元的全细胞记录

摘要

在大鼠下丘脑薄片标本上对52例视上核神经元进行了全细胞膜片籍记录。膜被动及主动电生理参数测量如下:静息电位,59士8 mV;输入阻抗,535士129 MQ;时间常数,32士9 ms;动作电位幅度,99士11 mV;超射值,37士13 mV (n=39)。大多数神经元在接受去极化刺激时出现明显的慢后超极化电位或电流。我们发现,在电压籍状态下几乎所有的视上核神经元均接受兴奋性和/或抑制性突触传入(n=13)。药理学实验表明,兴奋性突触后电流是由non-NMDA亚型谷氨酸受体介导,而抑制性突触后电流由GABA、受体介导。

WHOLE-CELL RECORDINGS OF THE SUPRAOPTIC NUCLEUS NEURONS FROM RAT HYPOTHALAMIC SLICES IN VITRO

Abstract

Fifty-two supraoptic nucleus neurons in rat slice preparations were studied using whole cell patch-clamp technique. The mean passive and active membrane properties were measured as follows:resting membrane potential,59士8 mV;input resistance,535士129 MQ;time constant,32士9 ms;amplitude of the action potentials,99士11mV;overshoot,37士13 mV(,,=39).Most of these neurons showed a prominent slow after- hyperpolarization potential or current in response to depolarizing pulse. In votageclamp condition,it was found that virtually all supraoptic neurons (n= 13) were invad ed by spontaneous synaptic inputs. Pharmacological experiments showed that the excitatory postsynaptic currents (EPSCs) were mediated by non-NMDA glutamate receptors,whereas inhibitory postsynaptic currents (IPSCs) by GABAA receptors.
