海马内注射脑啡肽对细胞免疫功能及脑内IL-l#alpha#基因表达的影响

摘要


文研究了海马内微量注射脑啡肽对大鼠细胞免疫功能及海马内白细胞介素一la (IL-la)基因表达的影响。结果发现:(1)双侧海马内微量注射甲硫一脑啡肽或亮一脑啡肽各1 u1 (18 mmol/L)能明显增强Con A刺激的脾淋巴细胞增殖活性,而双侧海马内微注射脂多糖各1 }t1 (50 ng/PI)则起抑制作用。切除双侧肾上腺后,海马内注射脑啡肽仍可增强脾淋巴细胞增殖反应。低浓度亮一及甲硫一脑啡肽(10-'0, 10-'1 mol/L)直接作用于体外培养的大鼠脾淋巴细胞,也明显增强Con A刺激的细胞增殖活性。(2)双侧海马内微量注射脂多糖各1 u1 (50 ng/wl),于90 min后用逆转录一聚合酶链反应技术检测到海马结构内IL-1。基因表达,而海马内甲硫或亮脑啡肽注射30 min后再给予脂多糖,则未检测到IL- l a cDNA的扩增条带。上述结果表明,海马结构对机体免疫功能的调节中脑啡肽起着重要作用,其机制可能与抑制海马结构IL-la基因表达有关。

EFFECT OF INTRAHIPPOCAMPAL MICROINJECTION OF ENKEPHALIN ON CELLULAR IMMUNE FUNCTION AND BRAIN IL- I #alpha# GENE EXPERSSION IN RAT

Abstract


The effect of intrahippocampal m.icroinjection of enkephalin on cellular immune function and hippocampal 1L- 1 u gene expression was studied in rats. The results showed that:(1)The proliferation activity of splenie lymphocytes stimulated by Con A was significantly increased with microinjection of Leu一enkephalin I p1(18 mmol/L) or Met-enkephalin I ucl(18 mmol/L)into bilateral hippocampus,but was inhibited by Lipopolysaccharide 1 41(50 ng/ul).The proliferative response of splenic lymphocytes to Leu-enkephalin still persisted after bilateral adrenalectomy. Low doses of Leu- or Met-enkephalin(10’。,10一‘工mol/L) were also capable of causing proliferative activi-
ty of the Con A- stimulated cultured splenic lymphocytes:、ritro.(2)Using RT-PCR technique,IL--1 a gene expression was detected in hippocampus 90 min after bilateral irrtrahippocampal injection of LPS. The LPS-induced IL- l u. gene expression in hippocarrr pus could no longer be detected 30 min after microinjection of Met一or Leu一enkephalin into hippocampus. The above results suggested that intrahippocampal enkephalin might play an important role in neur<〕一itnmunomodulation by enhancing the inhibition of 1L-lc. gene expression in hippocampal formation.
