PAF对肺内小动脉平滑肌细胞TxA_(2), PGI_(2) 及Cat~( + )-ATPase的影响

摘要
本工作采用分离培养家兔肺内小动脉平滑肌细胞(PASMCs ),观察了外源性血小板活化因子(platelet activating factor, PAF), BN52021 (PAF受体拮抗剂)、A引噪美辛、维拉帕米对PASMCs产生血栓素A2 (TxA2)、前列环素((PG12)及对细胞膜Cal+-ATPase活力的影响。结果表明:(1)基础状态下PASMCs存在花生四烯酸(AA)代谢。(2)外源性PAF通过受体后途径激活环加氧酶促进AA代谢致TxA2及PG工:增加,TxA2/PGI:比值无明显变化。(3)外源性PAF能直接抑制Ca l+-ATPase活力。(4)维拉帕米可逆转PAF抑制PASMCs膜Ca' -ATPase活力的效应。

Abstract
The effects of exogenous platelet activating factor (PAF),BN52021 (an antagonist of PAF receptor),indomethacin and verapamil on the production of TxA2,PG I2and activity of Cat一ATPase were investigated in rabbit intra-pulmonary smooth muscle cells (PASMCs).The results were as follows:(1)Under basic condition,active AA metablism was present in PASMCs.(2)Singnificant increase of TxA2 and PGI2 but not their ratio may result from binding of PAF receptor and activiton of cyclooxgenase.(3)Activity of Cat一ATPase could be inhibited by exogenous PAF.(4)The inhibitory effect of PAF on PASMCs Cat十一ATPase could be reversed by verapamil.
