三氯化铝对大鼠海马CA1区锥体细胞瞬间外向钾电流和延迟整流钾电流的影响

摘要
采用全细胞膜片钳技术,研究了三氯化铝(AlCl_(3))对急性分离的大鼠海马CA1区神经元钾通道的影响。结果表明,AlCl_(3)对钾电流有明显的抑制作用,具有一定的浓度依赖性,1000#mu#mol/L AlCl_(3)可改变I_(A)和I_(K)激活曲线和失活曲线的V_(h)和k值,使钾电流激活曲线右移,使失活曲线左移。这些结果表明AlCl_(3)对大鼠海马CA1区神经元K~(+)通道有抑制作用,它可能是铝引起中枢神经系统损伤的机制之一。

Abstract
The effects of aluminum chloride (AlCl_(3)) on the transient outward potassium and delayed rectifier K~(+) current in hippocampal CA1 neurons of rats were studied by the whole--cell patch clamp technique. It was found that AlCl_(3) reduced the transient outward potassium current and delayed rectifer K~(+) current in a dose-dependent manner. 1000 #mu#mol/L AlCl_(3) resulted in change in voltage and slope of the half-activation and the half-inactivation of I_(A) and I_(K). These results imply that AlCl_(3) may damage potassium channel of the hippocampal CA1 neurons from rats and this may be related to the mechanism of the damage to the central nervous system by aluminum.
