稻瘟病菌侵染诱导的水稻早期反应基因的cDNA片段克隆与序列分析

摘要

以稻瘟病菌感染水稻,利用mRNA差异显示技术分离了稻瘟病菌侵染诱导的水稻早期反应基因ER1(early responsive gene)的cDNA片段。Northern blot分析表明,ER1基因在稻瘟病菌侵染水稻叶片6h后开始表达,8h最强,10-12h开始减弱,16h消失。Southern blot分析表明,ER1基因属于水稻基因组。对ER1基因片段(219bp)进行了克隆和序列分析。经查询,在GenBank中没有与ER1同源的基因序列。

Cloning and sequencing of cDNA fragment of early responsive gene of rice induced by blast fungus Magnaporthe grisea

Abstract

cDNA fragment of early responsive gene (ER1) of rice (Oryza sativa L. subsp. japonica No.4) induced by blast fungus Magnaporthe grisea was isolated using mRNA differential display technique. Northern blot analysis showed that the expression of ER1 gene was induced at an early stage of blast infection. It started to express at 6h, strongly expressed at 8h, weakly expressed at 10h and 12h, and stopped expressed at 16h after blast infection. Southern blot shows that ER1 is one of genes of rice. ER1 gene has been cloned, DNA sequence analysis (219bp) showed no homologue in GenBank of ER1 gene.
