Zn~(2+)对爪蟾卵母细胞表达鲫鱼脑GABA受体的调制作用

摘要

爪蟾卵母细胞注射鲫鱼脑mRNA后表达的GABA受体中约85%为GABA_(A)受体。约15%的成分为GABA_(C)受体。利用双电极电压箝方法结合药物灌流研究了Zn~(2+)对这两型受体的作用。观察到Zn~(2+)对它们的调制都是抑制性的、可逆的。然而,与GABA_(A)受体相比,GABA_(C)受体对Zn~(2+)不太敏感。其中Zn~(2+)抑制鲫鱼脑GABA_(A)受体的IC_(50)=48.4±10.1#mu#mol/L,而它调制GABA_(C)受体的IC_(50)却高达255.6±21.5#mu#mol/L。

Zinc modulation of GABA receptors expressed in Xenopus oocytes injected with carp brain Poly(A)~(+) RNA

Abstract

Two types of GABA receptors, GABA_(A) receptors (approximately 85%) and GABA_(C)-like receptors (approximately 15%), were expressed in Xenopus oocytes after injection of Poly(A)~(+) RNA with carp (Carassius carassius) brain. Using voltage-clamp technique and pharmacological methods, the effects of zinc on the two receptors were studied. It was found that zinc modulation of both GABA_(A) and GABA_(C)-like receptors by zinc was inhibitory and reversible with the former being stronger than the latter as indicated by a value of IC_(50) of 48.4±10.1#mu#mol/L against 255.6±21.5#mu#mol/L.
