自发性高血压大鼠血管平滑肌细胞的精氨酸转运和脂质体对转运的影响

摘要

自发性高血压大鼠(SHR)的主动脉平滑肌细胞(SMC)转运L-精氨酸(L-Arg)明显低于WKY大鼠。SHR的SMC高和低亲合转运的最大转运速率(V_(max))分别为WKY大鼠的48%(P<0.01)和49%(P<0.01), 而米氏常数(K_(m))却无明显变化(P>0.05)。TNF #alpha#诱导SHR的SMC L-Arg转运速率增加的幅度明显低于WKY大鼠(P<0.01)。用包裹L-Arg的脂质体孵育SMC,细胞L-Arg摄入较游离L-Arg孵育增加数倍到数十倍,其转运率在SHR与WKY大鼠间相近,差异无显著性(P>0.05), 两种大鼠的SMC对脂质体-L-Arg的转运都不受TNF #alpha#预孵育的影响。实验结果表明,SHR的SMC存在有L-Arg的细胞转运障碍;用脂质体作为载体可显著提高SMC对L-Arg的转运效率。用脂质体包裹L-Arg治疗高血压可能具有潜在临床应用价值。

Abstract

There exists a functional disturbance in L-Arg transport in the SMC of SHR, but the L-Arg transport in SMC can be obviously enhanced when liposome is used as L-Arg carrier. Thus, it appears that liposome-L-Arg may have clinical perspective in the treatment of hypertension.
