摘要
Na_(2)SO_(3)对CF_(1)-ATPase活力的促进作用与酶所处状态有关。CF_(0)降低CF_(1)对Na_(2)SO_(3)的亲和力和Na_(2)SO_(3)促进的最大反应速率。在Na_(2)SO_(3)作用下,膜上CF_(1)-ATPase的活化能高于游离的。膜上和游离CF_(1)-ATPase的#gamma#亚基上二硫键的还原可以提高NA_(2)SO_(3)对酶活力的促进作用。Na_(2)SO_(3)对甲醇活化的CF_(1)-ATPase活力的促进作用只有在甲醇活化的亚适浓度下才能充分表现出来。Na_(2)SO_(3)对Mg~(2+)抑制的解除作用因CF_(1)-ATPase处于不同活化状态而不同。

Study on the stimulative effect of sulfite on the activity of CF_(1)-ATPase in different states

Abstract
The effects of sulfite on CF_(1)-ATPase in different methods were studied. Sulfite stimulate stimulated the activity of heat/DTT-activated soluble and thylakoid membrane-bound CF_(1)-ATPase activity was weaker. The affinity of heat/DTT-activated membrane-bound CF_(1)-ATPase for sulfite and the maximal activity of heat/DTT-activated membrane-bound CF_(1)-ATPase stimulated by sulfite were lower than those of soluble CF_(1)-ATPase. In the presence of sulfite, the activation energy of thylakoid membrane-bound CF_(1)-ATPase was higher than that of soluble CF_(1)-ATPase. These results indicate that the catalytic function of CF_(1) is controlled by membrane-bound CF_(0). The reduction of disulfide bond in γ subunit could further increase the stimulative effect of sulfite on heat- or methanol-activated soluble and thylakoid-membrane-bound CF_(1)-ATPase. The stimulative effect of sulfite on the methanol-activated soluble and thylakoid membrane-bound CF_(1)-ATPase was dependent on the methanol concentration in the reaction medium. Its stimulative effect was more pronounced at sub-optimal methanol concentrations than at the optimum concentration. The relieving effect of sulfite on the inhibiton of the methanol-activated CF_(1)-ATPase activity by Mg~(2+) was different from that of the heat/DTT-activated CF_(1)-ATPase. Why sulfite has different effects in relieving the inhibition of the activity of differently activated CF_(1)-ATPase by Mg~(2+) needs further study.
