向日葵下胚轴原生质体高频率愈伤组织形成与根发生

摘要
向日葵籽苗下胚轴原生质体,培养在含有BA 0.5 mg/L、2, 4-D 0.5 mg/L、NAA 0.1 mg/L和葡萄糖0.55 mg/L的改良Kao培养基中,24-28 h后,原生质体开始分裂。包埋在琼脂糖0.6%中的原生质体,培养5d后,分裂频率达95%以上。生长旺盛的小愈伤组织转移到含有2ip 0.1 mg/L、IAA 0.1 mg/L、腺嘌呤40 mg/L和GA_(3) 0.01 mg/L的Thompson液体培养基上13d后,原生质体诱导的少数愈伤组织发生根分化。
High frequency callus formation and limited rhizogenesis with hypocotyl protoplasts of Helianthus annuus L.

Abstract
Protoplasts isolated from hypocotyls of Helianthus annuus seedlings grown in the dark were cultured on modified medium after Kao (1982) containing BA 0.5 mg/L, 2, 4-D 0.5 mg/L, NAA 0.1 mg/L and glucose 0.55 mol/L as the sole source of carbohydrates. Protoplasts started to divide after 24-28 h of culture and after five days 95% of protoplasts immobilized in 0.6% agarose had divided at least once. Rapidly growing microcalli were transferred to liquid medium after Thompson et al. (1986) containing 2-iP 0.1 mg/L, IAA 0.01 mg/L, adenine 40 mg/L and GA_(3) 0.01 mg/L. A few calli derived from protoplasts produced a root each after 13 days of culture.
