Abstract
Capsule has restricted the further study on muscle spindle function and the involved mechanism. The aim of this study was to explore the isolation method of intrafusal fibres from the isolated rat muscle spindle. Intrafusal fibres were harvested from muscle spindle of soleus muscle in rats treated by neutrase -collagenase digesting. The isolated intrafusal fibres were identified under a microscope,and patch clamp was used to record resting potential. A variety of incubation mediums have been used to find out some survival medium of intrafusal fibers in vitro. The results indicated that the intrafusal fibres could survive for 1~2 h in the Leiboviz`s 15 Medium. The resting potential of intrafusal fibres obtained from patch clamp recording was -45.3±5.1 mV. The results are consistent with others obtained in vivo muscle spindle from cats and frogs. It is suggested that the intrafusal fibres are in good condition and enough to be used in experiments involving patch clamp. This study provides us a new approach in better understanding of muscle spindle activities and the involved mechanism.
