Abstract
Methanol extracts of the fresh skin of China amphibians (Bombilta orioatalis)
caused contraction of the smooth muscles of rat in vitro. The crude extracts were isolated by column chromatography of alkaline alumina, and a bioactive fraction was obtained from the 80%ethanol eluate. Submitting the fraction on Sephadex G_(-12) column.we found that the early eluate C_(-12),showing more bioactivity, was inactivated by hydrolysis with chymotrypsin, but not completely antagonized by cyproheptadine. C_(-12) was further analyzed by reversed phase-high performance liquid chromatography (RP-HPLC).The elution time of C_(-12-h) peak was the same as that of bombesin, and these two peptides had the same amino acid composition. These experimental results suggest that C_(-12-h) peptide is most probably bombesin.
