A preliminary observation on the hemopoietic inductive effect of marrow stroma

Abstract

The adherent cell layer formed by cultivating bone marrow cell in a liquid
system in dishes was taken as a stromal layer (SL) for studying the interaction of the hemopoietic cells and the stroma. The granulocyte-macrophage progenitor
cells (CFU-C) culture system in agar was overplated directly onto the human and mouse marrow SL which has been incubated for 1 to 3 weeks. In comparison with the CFU-C grown without SL as a control, the CFU-C grown on the first week SL were 51.07±3.540o (human) and 27.30±4.6800 (mouse) respectively; on the second week SL, the growth of CFU-C was practically the same; while the sum of the CFU-C colonies overlayed onto the third week SL increased up to 150.70±17.63%(human), and 146.89±10.01% (mouse).The results showed that SL affected granulopoiesis from inhibition to stimulation in a period of 3 weeks. This affection was probably mediated by the colony stimulating factor.
