ALTERATION OF CARDIAC #alpha#_(l)-ADRENERGIC RECEPTOR SUBTYPES IN CARDIAC INSUFFICIENT RAT

Abstract
The cardiac insufficiency was produced by simultaneous constriction of abdominal aortae and renal artery in rat. Radioligand binding and reverse transcription- pol),merase chain reaction (RT-PCR) were used respectively to determine the alterations of cardiac a,-adrenergic receptor (a,-AR) subtypes at protein level and gene transcription level. The Scatchard analysis of 1zsI_BE 2254 saturation curves showed that in cardiac insufficient rat hearts the KD values (229士32 vs 195士15 pmol/L) and the Bm$X vat ues(102士12vs96士17 fmol/mg) were not significantly different from those of control hearts. In the insufficient rat hearts,the inhibition of a,一AR specific binding by
WB 4101 showed that the high affinity sites (a,A+a,) were increased from the 22士5%of the control to 51士7%(PLO. 01).The a,A-AR mRNA level was increased,
a,B-AR mRNA level decreased while a,D-AR unchanged. The results suggest that in cardiac insufficient rat hearts the subtype composition is altered as shown by increased a,A-AR,decreased a1B-AR and unchanged a,D-AR,although the total amount of a,-AR isunchanged.
