Abstract
Effects of gold particle amount pe bombardment (30, 100, 250, 500 #mu#g) on transient and stable transformation of immature wheat embryos, were studied. And 500 #mu#g gold particle per bombardment is superior to others both in GUS transient expression and transgenic plant production. PCR and Southern analyses of T_(0) and T_(1) plants confirmed the integration of foreign bar gene fragments into wheat genomes. T_(1) plants smeared with L-PPT 200 mg/L showed a low level of resistance to this herbicide. The comparison of phenotypic character of T_(0), T_(1), T_(2) and their control plants showed that the phenotype abnormality of T_(0) plants is only a temporary phenomenon due to physiological and cultivated conditions, and the plants will recover from this abnormality partially or fully in the following progenes.
