Effect of ET-1 on intracellular free calcium in cultured neonatal myocardial cells

Abstract
In this present study, the effects of ET-1 on intracellular free calcium concentration ([Ca~(2+)]_(i)) and the underlying mechanisms were investigated in cultured neonatal rat myocardail cells loaded with fura-2/AM. The results are as follows. ET-1 induced an increase of [Ca~(2+)]_(i) in a dose-dependent manner, which consisted of transient and sustained phase. BQ123, a selective ET_(A) receptor antagonist, blocked the ET-1 induced [Ca~(2+)]_(i) responses, suggesting that these responses were mediated by ET_(A) receptors.
