Effects of H_(2)O_(2), ·OH and their scavengers on the activities of H~(+)-transport in the tonoplast vesicles isolated from barley leaves

Abstract

Both hydrolytic and H~(+)-transport activities of H~(+)-ATPase and H~(+)-PPase in the tonoplast vesicles isolated from barley leaves were lowered by H_(2)O_(2) and ·OH treatments. The hydrolytic activity of H~(+)-PPase was more sensitive to H_(2)O_(2) than that of H~(+)-ATPase. The depressed hydrolytic and H~(+)-transport activities of these two enzymes were recovered significantly by the addition of AsA, mannitol at the same concentration as H_(2)O_(2) and ·OH. K_(m) value of H~(+)-ATPase was increased and V_(max) of H~(+)-PPase decreased by treatments with H_(2)O_(2) and ·OH.
