Studies on the morphogenesis in different cultures of leaf explants of sense and antisense adp transgenic tobacco

Abstract
The leaf explants of tobacco (Nicotiana tobacum, SR1) and transgenic tobacco lines expressing foreign auxin binding protein (ABP) gene in sense (SE2) and antisense (AS3) orientation were cultured on MS media with 6-BA 2mg/L or 1mg/L and different concentrations of NAA for bud formation. At lower concentrations of NAA, more adventitious buds were formed in SE2 than that in SR1 and AS3, and the sensitivity of SE2 to auxin was increased. Feweer buds were formed in SE2 and SR1 than in AS3 at high concentrations of NAA, and the sensitivity of AS3 to auxin was decreased. When the leaf explants were cultured on media with polar transport inhibitor of auxin (HFCA), the differentiation of adventitious buds was inhibited.
