Transcription of RAB genes in guard cells of Vicia faba

Abstract
Guard cells of V. faba were prepared by conicating epidermal peels and treated with ABA, mannitol and Ca~(2+). RNAs extracted from leaf tissue and guard cells were hybridized with RNA probes of several representative RAB genes. An RNA with a sequence cognated to pea dehydrin (psdhn 1) was detected in water-stressed leaves and in the guard cells treated with ABA for 1 and 6 h. No RNA cognate to osmotin gene was detected in either tissue preparation. No difference was found between the transcripts of RAB genes from whole leaves and those from guard cells. ABA, but not Ca~(2+), was required for the formation of sequence cognated to psdhn1 in guard cells. Exogenous osmotan did not promote the formation of the RNA sequence.
