Studies on the cell type compositions of embryogenic callus of foxtail millet by using the micro-optic density image analysis system

Abstract
The physiological state of embryogenic cell lines is one of the most unstable factors, which often lowers the efficiency of protoplast culture system of cereal crops. In this report, preliminary results of studies on the glutamate dehy-drogenase (GDH) cell type compositions by using cytochemistry and microoptic density image analysis system were presented. The results showed that it was more accurate to assess the physiological states of embryogenic cell lines by using single cell population; the GDH cell type compositions of embryogenic cell lines could be used as a measure of the suitability of the cell lines for protoplast culture; the GDH cell type compositions could also be applied to the study of protoplast culture and tissue culture.
