Nuclear protein binding sites in 5^-upstream region of the rice waxy gene

Abstract

A 5^-upstream region (from-2 120 bp to transcription start site of this gene) was sequenced, fifteen small fragments were obtained and subcloned from the 2 120 bp fragment by different restriction enzymes. These fragments were labelled with ~(32)P and used as probes in gel retardation and cognate DNA competition assays. The results showed that HinfIa-2, HinfIc-1, HinfIc-2, HinfId, RsaIa (which overlaps with HinfIa and HinfIc) and RsaIe could interact with nuclear protein factors. In addition, HinfIa-2, HinfIc-1, HinfIc-2, HinfId and RsaIe DNA fragments could also interact with the proteins extracted from rice pollen. All these results demonstrate that at least five nuclear protein binding sites are located in the 5^ upstream region of rice waxy gene.
