High frequency callus formation and limited rhizogenesis with hypocotyl protoplasts of Helianthus annuus L.

Abstract

Protoplasts isolated from hypocotyls of Helianthus annuus seedlings grown in the dark were cultured on modified medium after Kao (1982) containing BA 0.5 mg/L, 2, 4-D 0.5 mg/L, NAA 0.1 mg/L and glucose 0.55 mol/L as the sole source of carbohydrates. Protoplasts started to divide after 24-28 h of culture and after five days 95% of protoplasts immobilized in 0.6% agarose had divided at least once. Rapidly growing microcalli were transferred to liquid medium after Thompson et al. (1986) containing 2-iP 0.1 mg/L, IAA 0.01 mg/L, adenine 40 mg/L and GA_(3) 0.01 mg/L. A few calli derived from protoplasts produced a root each after 13 days of culture.
