Oxidative burst in tomato cell culture induced by the elicitor from Verticillium albo-atrum

Abstract

The oxidative burst in two near isogenic tomato cell cutures induced by the elicitor from V.albo-atrum was investigated. The oxidative bursts of resistant and suceptible cell lines in Gamborg^s B_(5)/2% sucrose or in 2% sucrose only were compared Resistant cells showed a much higher level of reactive oxygen 20 min after the elicitor was added in two media.Cell age growth density,shaking,concentration of the elicitor,some oxidase and cathions etc. affected the peak value of oxidative burst in resistant tomato cell cutures. However, the pattern of the time course of oxidative burst was the same.The reactive oxygen species were increased rapidly at about 10 min and up to its peak at 20 min.There was always a difference in oxidative burst between the two near isogenic tomato cell tultures and the relative light units in resistant cell culture exhibited by luminol chemiluminscence was 1000 to 1000 times that in the susceptible one.The dependence of oxidative burst on concentration can be expressed by a sigmoid curve.
