Comparison between the properties of two forms of phosphofructokinase from spinach leaves

Abstract
Two forms of phosphofructokinase were separated by chromatography on a FPLC Sephacryl S-300 column from spinach leaf homogenate. The molecular weight of the large form was over 2000kD and that of the small form was 300kD. The large form was activated by inorganic phosphate, 3-PGA or citrate, but its activity was seriously inhibited by PEP. The inhibition could be relieved by Pi to some degree. The kinetic constants of the large form and the small form were: Mg~(2+) was required for the activity of both forms of PFK. The activity of large form was inhibited by a concentration of Mg~(2+) higher than 0.5mmol/L, while that of the small form was not affected.
