Immunogold electron-microscopy localization of abscisic acid in mesophyll cells of Vicia faba L.

Abstract

With immunogold electron microscopy the study on the localization of abscisic acid (ABA) in mesophyll cell of Vicia faba has been undertaken. The results showed that, in thin sections treated with ABA antibody, chloroplasts, cytosolic compartments and the nuclei were labelled by gold particles, but chloroplasts were labelled by more gold particles. However, no gold particles were found in cell wall and vacuoles. Treating the sections with trypsin before immunostaining could increase the gold particle density substantially. The sections with no EDC fixation or treated with preimmune rabbit serum were devoid of gold particle label.
