Characterization of phycobilisome and subunit composition of allophycyanin-APC 660 nm from the thermophilic cyanobacterium Myxosarcina concinna Printz

Abstract

Three allophycocyanins, two phycocyanins and one kind of red protein was separated from the intact PBS by PAA 4%~30% gradient gel electrophoresis. Only one easily dissociable C-phycoyanin and one relatively stable allophycocyanin (APC 660 nm) were obtained by using preparative PAGE and hydroxylapatite column chromatography. The fluorescence emission spectrum of the APC 660 nm at room temperature exhibited the maximum at 660nm and its fluorescence life time was shown to be 2.063ns. The molecular weights of the APC 660nm were found to be 140kD and 147kD, The APC 660nm contained two light and two heavy subunits having molecular weights of 16000(#alpha#), 17 000(#alpha#^), 18 400(#beta#), and 19 000(#beta#^), respectively. The subunit composition of APC 660 nm deduced from these results would be (#alpha##alpha#^ #beta##beta#^)_(2), which was different from the complex(#alpha#^ #alpha#_(2)#beta#_(2)#beta#^)Lc~(10) for APC 664 nm from thermophilic cyanobacterium Mastigocladus laminosus and (#alpha##beta#)_(3)for APC 660 nm from common blue-green algae.
