Preparation of ~(125)I-brassinosterone and its biological activity

Abstract
Brassinosterone, a precursor in the synthesis of brassinolide, exhibitedmore than 70% of the biological activity of brassinolide in rice test.Brassino-oxime, a product of condensation of brassinosterone withcarboxymethoxylamine hydrochloride, showed 60% of that of brassinolide in thesame test system at 10 #mu#mol/L concentration. In cucumber hypocotyl test,the brassion-oxime(2#mu#mol/L)promoted elongation of cucumber hypocotyl by 20%as compared with control. The brassino-oxime was found to be competitive withbrassinolide or brassinosterone. Combination of iodohistamine withbrassion-oxime by Chloramine-T method formed the ~(125)I-labelled compoundwhich was used for cucumber and rice tests. The results showed that the Rf0.93 fraction of chromatography was effective in promoting both elongation andinclination significantly. Its specific radioactivity was 73.8 GBq/mmol.
